Notre expertise en RMN pourra \u00eatre utilis\u00e9e en amont ou en aval du criblage pour valider et\/ou contr\u00f4ler vos chimioth\u00e8ques et d\u00e9terminer la solubilit\u00e9 des STD, WaterLOGSY) avec un ligand connu ou en l\u2019absence de comp\u00e9titeur par une exp\u00e9rience de cartographie des d\u00e9placements chimiques de la prot\u00e9ine en pr\u00e9sence du ligand. Outre l\u2019identification du site d\u2019interaction, cette derni\u00e8re approche permet la construction d\u2019un mod\u00e8le avec une prot\u00e9ine, soit pour l\u2019identification de partenaires physiologiques soit pour l\u2019identification de touches dans la cadre d\u2019un criblage de d\u2019interactions avec les r\u00e9gions hydrophobes des prot\u00e9ines. Le d\u00e9pliement de la prot\u00e9ine, et l\u2019exposition des parties hydrophobes, donnent alors naissance \u00e0 un motif caract\u00e9ristique de la fluorescence en fonction de la temp\u00e9rature<strong>.<span>\u00a0 <\/span><\/strong><span>\u00a0<\/span><\/p><p>Les applications concernent non seulement le criblage de ligands mais \u00e9galement l\u2019identification de conditions optimales pour la purification et le stockage des prot\u00e9ines et de leurs partenaires pour faciliter la cristallisation. Du fait du peu de mat\u00e9riel prot\u00e9ique utilis\u00e9, du d\u00e9bit relativement \u00e9lev\u00e9 de la m\u00e9thode et de la large gamme d\u2019affinit\u00e9 couverte, la fluorim\u00e9trie diff\u00e9rentielle \u00e0 balayage est un outil pr\u00e9cieux et parfaitement adapt\u00e9 pour le criblage par fragment.<\/p><\/div><\/div><\/div><section class=\"elementor-element Elle peut prendre differentes formes allant de la <\/span><strong>prestation de service \u00e0 la collaboration scientifique. <\/strong><span>Une <\/span><strong>\u00e9tude de faisabilit\u00e9<\/strong><span> est souvent n\u00e9cessaire.\u00a0<\/span><\/p><p>La \u00a0DSF (fluorim\u00e9trie diff\u00e9rentielle \u00e0 balayage) est une m\u00e9thode tr\u00e8s rapide permettant de cribler jusqu\u2019\u00e0 1000 compos\u00e9s en 2 jours sur la base d\u2019une interaction directe entre la cible et le ligand.<\/p><p>La spectroscopie RMN permet de cribler rapidement (en moins d\u2019une semaine), une librairie de taille \u00e9quivalente. <span>\u00a0<\/span>Il est en plus possible de valider le site de fixation du ligand en utilisant par exemple un comp\u00e9titeur d\u00e9j\u00e0 connu.<\/p><p>Lorsque les conditions de cristallisation de la cible sont connues, la cristallographie aux rayons X peut \u00eatre utilis\u00e9e comme m\u00e9thode de criblage. Elle est moins rapide que la RMN mais pr\u00e9sente l\u2019avantage de fournir en m\u00eame temps une identification du site d\u2019interaction et une compr\u00e9hension du mode d\u2019interaction.<\/p><p><strong>Une \u00e9tude ambitieuse de d\u00e9couverte de ligands fait g\u00e9n\u00e9ralement appel aux trois approches selon une strat\u00e9gie en trois \u00e9tapes successives: criblage initial (ex.
