Ces m\u00e9thodes permettent notamment de mesurer la solubilit\u00e9, la stabilit\u00e9, la masse molaire ainsi que l\u2019\u00e9tat de polydispersit\u00e9 et d\u2019oligom\u00e9risation des macromol\u00e9cules et leurs complexes.<\/p>\n<p>Ces \u00e9quipements peuvent notamment \u00eatre utilis\u00e9s en amont d\u2019une <strong>\u00e9tude structurale<\/strong> ou d\u2019un <strong>criblage<\/strong> pour caract\u00e9riser les \u00e9chantillons prot\u00e9iques et v\u00e9rifier qu\u2019ils remplissent les conditions n\u00e9cessaires \u00e0 l\u2019\u00e9tude envisag\u00e9e.<\/p>\n<p><\/p>\n<p>Elles sont \u00e9galement utilis\u00e9es pour caract\u00e9riser en d\u00e9tail le <strong>mode d\u2019interaction<\/strong> de diff\u00e9rents types <strong>de<\/strong> <strong>complexes macromol\u00e9culaires (interaction prot\u00e9ine-prot\u00e9ine (PPI), ligands, acides nucl\u00e9iques)<\/strong> et suivre les changements conformationnels pouvant en r\u00e9sulter.<\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-750b7402 elementor-widget elementor-widget-heading\" data-id=\"750b7402\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"heading.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t<h5 class=\"elementor-heading-title elementor-size-default\">Pr\u00e9requis<\/h5>\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element elementor-element-51653ee5 elementor-widget elementor-widget-text-editor\" data-id=\"51653ee5\" data-element_type=\"widget\" data-e-type=\"widget\" data-widget_type=\"text-editor.default\">\n\t\t\t\t<div class=\"elementor-widget-container\">\n\t\t\t\t\t\t\t\t\t<p>Toute \u00e9tude biophysique requiert un \u00e9chantillon prot\u00e9ique pur chimiquement. Cette puret\u00e9 doit \u00eatre valid\u00e9e \u00e0 minima par analyse sur gel en conditions d\u00e9naturantes. Certaines m\u00e9thodes, comme le microcalorim\u00e9trie demandent en plus une puret\u00e9 oligom\u00e9rique.<\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t<div class=\"elementor-element diffusion statique de la lumi\u00e8re \u00e0 plusieurs angles (MALS), de l\u2019index de r\u00e9fraction (RI) et de l\u2019absorbance (UV) est une m\u00e9thode rapide et pr\u00e9cise pour d\u00e9terminer la masse molaire des prot\u00e9ines et des complexes en solution et l\u2019\u00e9tat d\u2019oligom\u00e9rie. Elle permet \u00e9galement de contr\u00f4ler l\u2019homog\u00e9n\u00e9it\u00e9 conformationnelle des \u00e9chantillons.<\/p>\n<div class=\"elementor-element elementor-element-38232d4 elementor-widget La technique n\u2019\u00e9tant pas destructive il est possible d\u2019effectuer des cin\u00e9tiques ou titrations pour suivre les changements conformationnels. Il est \u00e9galement possible de d\u00e9terminer la temp\u00e9rature de fusion (Tm) de la macromol\u00e9cule et ceci sans la pr\u00e9sence d\u2019un fluorophore, ce qui rend la m\u00e9thode CD compl\u00e9mentaire \u00e0 la DSF. La quantit\u00e9 et concentration d\u00e9pendent essentiellement de la cuvette utilis\u00e9e : de quelques \u00b5g\/ml pour la cuvette 500\u00b5L (chemin optique 1mm) \u00e0 quelques mg\/ml pour la cuvette sandwich (chemin optique 0.01mm). Les meilleurs tampons sont le phosphate, le borate et le Tris-HCl.
